Confocal laser scanning microscopy (CLSM) is one of the most important advances achieved during recent decades in the field of fluorescence imaging and is considered as an essential tool in biological research. Compared with electron microscopy, CLSM procures much poorer resolution but requires considerably less specimen preparation and is compatible with three-dimensional (3D) live imaging, enabling access to dynamic cellular and molecular processes.

Confocal microscopy offers several advantages over conventional optical microscopy, including controllable depth of field, the elimination of image degrading out-of-focus information, and the ability to collect serial optical sections from thick specimens. Laser Confocal Microscopes Olympus Laser Confocal Microscope offers superb image quality and accurate 3D measurement by non destructive observation method with advanced optical system. Its operation preparation is easy and no pre-process is necessary with your samples. Confocal microscopy, most frequently confocal laser scanning microscopy (CLSM) or laser confocal scanning microscopy (LCSM), is an optical imaging technique for increasing optical resolution and contrast of a micrograph by means of using a spatial pinhole to block out-of-focus light in image formation.[1] [Confocal laser scanning microscopy].

A confocal microscope creates a high optical resolution and high contrast image with a spatial pinhole, which blocks out-of-focus light in image formation. Laser scanning confocal microscopes employ a pair of pinhole apertures to limit the specimen focal plane to a confined volume approximately a micron in size. Relatively thick specimens can be imaged in successive volumes by acquiring a series of sections along the optical (z) axis of the microscope. Laser Scanning Confocal Microscopy Typically, confocal microscopy utilizes wet- immersion objectives with magnification of X60 – X100. Typical laser beam diameters on the objective are ¬6mm, with a resulting diffraction limited spot size of less than 1µm.

In confocal microscopy, laser light is focused into a very small spot and is scanned across the specimen to reconstitute a 2D image. This is in contrast to  Learn More About the Caliber I.D. Confocal laser scanning microscopy can be used for countless applications and research areas. Images produced by confocal laser scanning microscopy are especially well suited to assist physicians to perform screening examinations and diagnoses of skin cancer or early forms of skin cancer (carcinoma in situ). Perhaps the most significant advance in optical microscopy during the past decade has been the refinement of mainstream laser scanning confocal microscope (LSCM) techniques using improved synthetic fluorescent probes and genetically engineered proteins, a wider spectrum of laser light sources coupled to highly accurate acousto-optic tunable filter control, and the combination of more advanced software packages with modern high-performance computers. 2018-10-28 · Laser Scanning Microscopy Tutorial.

In short, very thin focal planes are imaged and these deliver high detail optical sections through the sample. If multiple sections at different depths are scanned, a 3D-reconstruction of the sample can be made.
